
This article describes the development of a flow-through (FT) polishing step using hydrophobic interaction chromatography (HIC) to remove mAb aggregates.
It includes:
– The use of HTPD plates to select the optimal resin as well as running conditions
– The use of a design of experiments (DoE) to optimize the running conditions
– A Monte Carlo simulation to identify the robust conditions to reach the objectives in aggregation removal and the monomer recovery.
